The 3BPlasmid DNA Purification kit is designed for a rapid and easy preparation of up to 20 μg of highly pure plasmid DNA from 0.5 to 2 ml bacterial suspension (minipreps). It is based on a modified alkaline lysis procedure followed by binding of the plasmid DNA to a silica membrane column in a comfortable spin format.
- Fresh cultured bacterial suspension.
- Frozen bacteria pellets.
Harvested bacteria are resuspended in RNase containing solutions and processed by alkaline/SDS lysis. The resulting lysate is neutralised, and the plasmid DNA is bound to a silica membrane in the binding column. This is followed by a centrifugation step to clear the lysate. All impurities are efficiently removed by washing and the ultra purepDNA is eluted in a small volume of elution buffer.
The obtained plasmid DNA is ready to use for a wide variety of different downstream applications: PCR, reaction enzyme digestion, labeling, cloning, in vitro transcription, and capillary sequencing.
